PBMC were activated by anti-CD3 and anti-CD28. Th17 differentiation was promoted by adding 10 ng/mL of TGF-β, 10 ng/mL of IL-23, 10 ng/mL of IL-6, 10 ng/mL of IL-1β, and 1 μg/mL of anti-IL4 and anti-IFNγ with (C-TSLP or P-TSLP), or without (C or P)125 ng/mL of TSLP. The concentrations of IL-17 in the culture supernatants were determined by enzyme-linked immunosorbent assay. C: control; P: patient.
